Hours of instruction:

Weekly: four hours of lectures + four hours of labwork (4+0+4)

Goals:

I. To introduce students to the fundamentals of retrosynthetic analysis of organic molecules; developing students’ ability to plan synthesis and apply familiar reactions in the syntheses of relatively simple target molecules.

II. To introduce students to the traditional and modern approaches in the field of the chemistry of heterocyclic compounds.

III. Introduction to medicinal chemistry and biological activity of some classes of heterocyclic compounds.

Outcome:

I. Ability to independently perform retrosynthetic analysis of some simple/moderately complex organic molecules.

II. Understanding structure, properties and reactivity of some classes of heterocyclic compounds.

III. Acquired knowledge of the properties which differentiate an interesting molecule from a drug.

Lectures:

Syllabus:

I. The elements of complexity of organic molecules. Basic concepts of retrosynthetic analysis: synthon, retron, transform. Synthonic approach. Systematic retrosynthetic analysis; different strategies of analysis. Transform-oriented strategies; types of transforms. The influence of functional groups, stereochemical elements, topology, symmetry of molecules and other structural elements on the course of retrosynthetic analysis. Selected examples of total syntheses.

II. Azoles: 1,2-azoles and 1,3-azoles. Benzo-1,2-azoles and benzo-1,3-azoles. Diazines: pyridazine, pyrimidine and pyrazine. Purines. Heterocyclic compounds with more than two heteroatoms. Methodological approach in the synthesis of biologically active heterocyclic compounds.

III. Biological activity. Importance and properties of a drug. Desirable properties of a drug candidate. The most important phases in drug discovery. ADMET, research and importance of a certain activity.
